Figure 5. Specific binding of IL-16 to CD9-expressing CHO cells. CHO cells transfected with pcDNA3 alone (B) or pcDNA3 containing a CD9 cDNA (A,C-D) were seeded in 24-well chamber slides. Three days later, the cultured cells were washed and preincubated with no Ab (A-B), isotype-matched mouse IgG (C), or anti-CD9 mAb (D). The binding of IL-16 to the transfectants was visualized using Alexa Fluor-488–conjugated IL-16 and was evaluated by fluorescence microscopy using a Leica Microsystems Diaplan fluorescence microscope and 25×/0.60 water immersion objective. Images were acquired using a Leica Microsystems DFC 480 digital camera and Leica Microsystems DFC Twain 6.1.1 software.
