Yield of tetramer-positive cells of 5 pp65495-503-specific CTLs that were purified by flow sorting or with anti-PE–coated paramagnetic beads. The number in the second column represents the number of CD8+ cells with which the culture was commenced. The numbers in the other columns indicate the total number of cells in culture (× 106); the percentage tetramer-positive cells is indicated in parentheses. One bead-purified pp65495-503-specific CTL lines (no. 5) was not expanded; the rapid expansion of line 2 failed probably due to technical problems during flow sorting, which increased the sorting time substantially and resulted in dead cells in the sorted population.
