Cells were sorted in numbers of 1, 2, and 5 to each well in Terasaki trays containing media supplemented with IL-1β, IL-3, IL-4, IL-7, SLF, TNF-α, and Flt3-L. In each population, 30 wells were plated with 1 cell, and 20 wells were plated with 2 and 5 cells. Sorted numbers were confirmed by examination 2 to 4 hours after plating. Cells were evaluated as described in “Materials and methods” on day 4 of culture. Total plating efficiency indicates percentage total plated wells with viable cells, and DC plating efficiency indicates percentage total wells with cells displaying DC phenotype. Average number of cells per well refers to all visible cells per plated wells. Average number of DC per well refers to all wells showing cells with DC phenotype.
