Total or magnetically depleted Lin− or B220− spleen cells were preincubated for 24 hours at a concentration of 107, 106, or 107cells/mL, respectively. Total and sorted cell populations were plated in a final volume of 1 mL MethoCult at concentrations ranging from 0.2 to 5 × 105 cells per culture dish and scored for colonies on day 7 or 8. Data represent means ± SEM from triplicate determinations of a typical experiment reproduced twice.
