MCF7 cells were transiently cotransfected for 48 hours with CMV-GFP and either CMV empty vector (−), CMV-p15, CMV-p21, or CMV-p27, as indicated. Expression of GFP was used to sort GFP-positive cells using a Becton Dickinson cell sorter. GFP-positive cells were then used for subsequent cell cycle analysis by determining DNA content by propidium iodide staining. Data were processed using the multicycle program (Phoenix Flow System). For comparison, cells treated for 48 hours with 1,25(OH)2D3 were also analyzed for DNA content (bottom line).
