Chimeric bone marrow samples were stained with various monoclonal antibodies specific against different human blood cell lineages and CD45 as a marker for human cells. The gated human CD45+ cells were analyzed for GFP expression and subsequently for expression of human CD19 (B lymphoid), CD33 (myeloid), or CD34. Percentages of GFP+ and GFP−lymphoid, myeloid, and CD34+ cells, respectively, are presented as percentages of the human GFP+ and GFP− CD45+ cell population.
Mice from both independent transplantation experiments were combined for the analysis. Statistical analysis was performed by applying the paired, 2-sided Student t test.

In 4 of 8 mice receiving transplants of HOXB4, the proportion of CD33+ cells in the GFP+ population was significantly lower than in the GFP− population: 8.3 ± 1.9 versus 22.3 ± 4.5, respectively; n = 4;P = .01.
